Abstract:

Regulation of chromatin landscape is intimately linked with DNA-templated processes such as transcription, replication, and DNA damage repair. Aberrant chromatin regulation can serve as an underlying cause of genetic disorders and cancers. The ATP-dependent chromatin remodeling complexes (remodelers) are multicomponent chromatin regulators that play critical roles in establishing and modulating genomic architecture. These megadalton-sized molecular machines are mutated in over 20% of malignancies, underscoring their functions in gene expression. It is known that remodelers can directly interact with multiple histone domains of the nucleosome to elicit their functions. However, the specific histone features that govern their binding and activities remain undefined. I will discuss how chemistry-based approaches can help elucidate the molecular interplays between chromatin and chromatin remodeling complexes by giving you three examples. First, I will describe a photocrosslinking-MS-based technology (‘photoscanning’) for profiling protein-histone interaction and its applications to identify unknown histone interacting domains of the ISWI and SWI/SNF remodelers. Next, I will discuss how cancer-associated histone mutations disrupt the directionality of chromatin remodeling based on remodeling assays with novel nucleosome substrates. Finally, I will discuss a high-throughput screening platform (termed DNA-barcoded nucleosome library) for studying the effect of histone PTMs on the binding and activity of the SWI/SNF remodelers.
