Abstract:

Gene promoter hypomethylation in some human cancers compared to their normal counterparts is discovered nearly 40 years ago. This loss of DNA methylation at promoter CpG dinucleotides is the first epigenetic abnormality to be identified in cancer cells. The hypomethylation of DNA can lead to transcription activation and mostly occur in oncogenes. In contradiction, a gain of methylation at the promoters of some tumor suppressor genes in a range of tumor types is also observed and initialize the gene deactivation. In short, DNA methylation regulates gene expression in tumor progression, but this is not the whole picture. The advancing of technology in next-generation sequencing and bio-molecular assays in recent years helps to reveal the leftover. In this first part of the presentation, I would like to describe the paradoxical role of DNA methylation in transcription modulation with other epigenetic marks of breast cancer to fully understand the underline mechanism. In the second part, I will focus on the work-flow of the development of DNA methylation as a biomarker for clinical diagnosis. First, I will show DNA methylation is a promising biomarker for early detection of breast cancer, then applying these markers on public genome-wide methylomes for computational prediction. Finally, the passed markers are validated in primary tissue/FFPE or solid biopsy and circulating cell-free DNA (ccfDNA) or liquid biopsy.
